A good solution for separations at high pH - ZORBAX Extend-C18 HPLC columns. Method validation kit with columns from three different batches/lots. The column presented here has an inner diameter of 4.6 mm, a length of 150 mm and a particle size of 5.0 µm.
Agilent ZORBAX Extend-C18 columns use a novel binding technology based on a two-tooth C18 structure. This allows separations to be performed with silica gel based columns at high pH. At high pH, non-charged, basic compounds do not interact with silica gel.
The result is separations with high efficiency, excellent peak shape and improved resolution. Separations at high pH are also popular if the compounds are then more stable or better soluble. Triethylamine, pyrrolidine, glycine, borate and ammonium hydroxide are suitable buffer substances for the mobile phase. Ammonium hydroxide is an excellent buffer for the mobile phase at pH 10.5. Compared to TFA-containing mobile phases, significantly better sensitivity can be achieved for LC/MS separations of peptides and small molecules.
The Extend C18 column is stable in the pH range 2-11.5 and provides a good peak shape for all types of substances.
