General Information about Normal Phase Chromatography

Normal phase chromatography uses the interaction of the analyte with polar functional groups on the surface of the stationary phase. These interactions are strongest when using nonpolar solutions as mobile phase. Furthermore normal phase chromatography is a very efficient separation method, cause of the large range of solvents that can be used to adust selectivity of the each separation. Through the high complexity this methods popularity decreased until today.
Under certain conditions long equilibrium time or problem with reproducability can occure caused by the sensitivity of this technique toward low concentrations of polar components in the mobile phase. Controling these problems provides normally better chromatograms as reversed phase chromatography caused by the lower viscosity  of common solvents.
Using normal phase chromatography means nonpolar mobile phases and polar stationary phases. The normal phases are made out of silica gels or aluminium oxide, which enable pure adsorption separation at the polar OH groups. The essential separation mechanism is based on varying adsorption of analyte molecules on the stationary phase´s surface.
